Anaplasmosis, borreliosis, rickettsiosis, and babesiosis, a class of tick-borne diseases, are of considerable medical, veterinary, and economic concern. Previous disease assessments in Belgium's animal populations have only offered a partial view of the prevalence of these illnesses, with the focus of these screenings limited to certain geographical areas, documented disease cases, or only a small number of tested specimens. To this end, we initiated the first comprehensive nationwide seroprevalence study encompassing Anaplasma species, specifically A. phagocytophilum, Borrelia species, and Rickettsia species. Belgian cattle are known to carry Babesia spp. We additionally screened questing ticks for the previously identified pathogens.
In each province, ELISA and IFAT tests were conducted on a representative serum sample set, sized proportionally to the cattle herd count. A fluorescence-based SYBR Green I test was utilized to examine the impact of combined therapy with diminazene aceturate (DA) and imidocarb dipropionate (ID) on the in vitro growth of various parasitic piroplasmids and Babesia microti in BALB/c mice. Our investigation focused on the structural similarities between routinely prescribed antibabesial drugs DA and ID, and newly identified antibabesial medications pyronaridine tetraphosphate, atovaquone, and clofazimine, employing atom pair fingerprints (APfp) for analysis. The Chou-Talalay approach facilitated the identification of interactions between the two drugs. The Celltac MEK-6450 computerized hematology analyzer served to identify hemolytic anemia in mice infected with B. microti and in those receiving either a single or combined therapy regimen, this procedure being performed every 96 hours. The APfp data suggests that DA and ID display the strongest structural overlap (MSS). Against the in vitro growth of Babesia bigemina, DA and ID demonstrated a synergistic effect, while against Babesia bovis, their interaction was additive. Concomitant treatment with low doses of DA (625 mg kg-1) and ID (85 mg kg-1) exhibited a greater inhibitory effect on B. microti growth (165%, 32%, and 45%) compared to the respective monotherapies of 25 mg kg-1 DA, 625 mg kg-1 DA, and 85 mg kg-1 ID. Within the blood, kidney, heart, and lung tissues of mice administered DA/ID, the presence of the B. microti small subunit rRNA gene was not established. The observed results point towards the possibility of DA/ID being a promising treatment option for bovine babesiosis. This joint application may surpass the problems of Babesia resistance and host toxicity resulting from using the full dosages of DA and ID.
